Autosampler Manual Injection Test:

- A manual injection was performed to test the autosampler’s functionality.
- The needle was observed dipping correctly into the sample solution.
- Injection confirmed that the autosampler syringe system was operational and capable of aspirating/dispensing liquid.


Flow Path and LC System Check:

- The LC pump and flow system were engaged and confirmed to be functional.
- Flow through the column and divert valve was verified, showing that the sample path to the mass spectrometer was intact.
- No leaks or pressure irregularities were observed, confirming the LC system’s stability.


Mass Spectrometer Verification:

- The TSQ Endura mass spectrometer was engaged and tested in operating mode.
- Signal response confirmed that the MS was detecting and recording ion events appropriately.
- This validated that the instrument was capable of receiving and processing analyte input from the LC system.

A 2 μL loop injection of a 500 fg/μL reserpine solution will produce a minimum signal-to-noise ratio of 25,000:1 for the transition of the protonated molecular ion at m/z 609.3 to the fragment ion at m/z 195.1 when operated in selected reaction monitoring (SRM) mode with Q1 and Q3 resolution both set to 0.7 Da FWHM.

Negative Electrospray (nESI)

A 2 μL loop injection of a 500 fg/uL chloramphenicol solution will produce a minimum signal-to-noise ratio of 80,000:1 for the transition of the deprotonated molecular ion at m/z 321.0 to the fragment ion at m/z 152.0 when operated in selected reaction monitoring mode (SRM) with Q1 and Q3 resolution set to 0.4 and 0.7 Da FWHM, respectively.
